Summary
Ten healthy lean women were studied during 3 days of isoenergetic diet, 3 days of carbohydrate overfeeding, and 3 days of fat overfeeding. Carbohydrate overfeeding raised plasma leptin by 28% and 24-hour energy expenditure by 7%, while basal metabolic rate and energy expended during physical activity were unchanged. Fat overfeeding changed neither leptin nor energy expenditure. Changes in leptin were unrelated to changes in energy expenditure, leading the authors to conclude that leptin does not mediate the metabolic stimulation of overfeeding.

Key findings
- 1
Three days of carbohydrate overfeeding raised plasma leptin by 28% and 24-hour energy expenditure by 7%.
- 2
Basal metabolic rate and energy expended during physical activity were unaffected by carbohydrate overfeeding.
- 3
Fat overfeeding significantly changed neither leptin nor energy expenditure; the response differs by macronutrient.
- 4
Changes in leptin were unrelated to changes in energy expenditure, so leptin cannot be said to mediate the metabolic stimulation.
- 5
Participants were 10 healthy lean women and the intervention lasted 3 days. Effects in people who are dieting or in a plateau, and the effect of a single day of overfeeding, were not tested.
